GtoPdb Ligand ID: 11306

Synonyms: (-)-borrelidin
Antimalarial Ligand
Comment: Borrelidin is a macrolide antibiotic originally isolated from Streptomyces. It is a noncompetitive inhibitor of both bacterial and eukaryotic threonyl-tRNA synthetases (also known as threonine--tRNA ligase), exhibiting antibacterial, antimalarial and antiangiogenic activities.

Guide to Malaria Pharmacology Comments
Borrelidin is cytotoxic, with a narrow therapeutic window, but a number of analogues have been developed that retain potent antimalarial activity and display improved selectivity [1,3].

Potential Target/Mechanism Of Action: the proposed molecular target is Plasmodium threonyl-tRNA synthetase (PfTRS).
