Comment: IL-27 is an IL-12 family cytokine. It is a heterodimeric cytokine that contains an α and a β subunit and is expressed by activated antigen presenting cells . IL-27 interaction with its receptor complex activates the JAK/STAT and p38 MAPK pathways.
|Production of IL-27 is driven by activation of signalling pathways (e.g. via TLR or TNF receptor activation) that impinge NF-κB/STAT1 transcription. IL-27 induces IFN-γ production by natural killer and T cells, producing a proinflammatory response. It is also involved in the balance between Th1 and Th2 immunity with the ability to antagonize Th1, Th2, Th9, and Th17 responses to control inflammation. IL-27 regulates both adaptive and innate immune responses. IIt inhibits the production of pro-inflammatory cytokines IL-1, IL-6 and IL-17, but induces the production of anti-inflammatory cytokine IL-10. The complexity of IL-27 immunobiology is reviewed in .
