Comment: Vepdegrestrant (ARV471) is a bifunctional compound that is estrogen receptor (ER) degrader (PROTAC) [1]. It uses a cereblon-based binder to direct ER protein degradation. ARV471 is orally bioavailable, and was proposed for potential to treat breast cancer. Vepdegrestrant was the first PROTAC to gain clinical approval (from the US FDA, May 2026).
