Comment: Tozorakimab (MEDI3506) is a high-affinity anti-IL-33 monoclonal antibody. It inhibits IL-33-induced signalling to produce an anti-inflammatory effect [2]. Tozorakimab blocks the actions of both reduced IL-33 (IL-33^red) and oxidized IL-33 (IL-33^ox) metabolites through their respective downstream pathways (ST2 for IL-33^red, RAGE/EGFR complex for IL-33^ox) [1,3-4].

Tozorakimab blocks IL-33 signalling via serum-stimulated 2 (ST2) and advanced glycation end products/epidermal growth factor receptor (RAGE/EGFR complex) signalling pathways, and mediates an anti-inflammatory outcome. It inhibits IL-33-mediated pro-inflammatory cytokine release from HUVECs, mast cells and peripheral blood mononuclear cells at picomolar concentrations [2].
