Synonyms: TYK2 PROTAC 15t [PMID: 40116635]
Compound class:
Synthetic organic
Comment: TYD-68 is a cereblon-recruiting PROTAC that promotes selective degradation of tyrosine kinase 2 (TYK2) [1]. It is a heterobifunctional molecule that binds to the allosteric pseudokinase domain (Janus homology 2: JH2) of TYK2 (using a BMS-986202 derivative) and to cereblon.

Bioactivity Comments |
The DC50 for TYD-68 vs. TYK2 protein is 0.42 nM [1]. A maximum 95% reduction in cellular TYK2 protein level is achieved using 10 nM compound and 10 hours incubation period. The parent molecule of the TYK2-binding moiety of TYD-68, BMS-986202, binds TYK2-JH2 with an IC50 of 0.19 nM. |
