In August 2013, Merrimack Pharmaceutical announced that they had been granted FDA orphan drug status for MM-111 for the treatment of advanced gastric and esophageal cancers. MM-111 reached Phase 2 clinical trial in HER2-positive gastric cancer. However, in early 2015, the maufacturer terminated development due to poor responses, including inferior progression-free survival in the experimental arm of the trial in metastatic HER2-positive gastric cancer patients.

MM-111 forms a complex with ErbB2 and ErbB3, which effectively inhibits ErbB3 signaling. This action underlies the antibody's antitumor activity in preclinical models [1].
