Comment: This is the lipopeptide version of EK-1 [1]. It has cholesterol fused at the C terminal via a short peptide linker and a flexible polyethylene glycol (PEG) spacer. Like EK-1 it acts as a viral fusion inhibitor that blocks infection by a range of human coronavirus pseudoviruses and live viruses, including SARS-Co-V and SARS-CoV-2. It is significantly more potent than EK-1, and is active in vitro and in vivo.
Bioactivity Comments
EK-1-C4 inhibits SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM respectively.
