Comment: GNE-4997 is a tetrahydroindazole-containing inhibitor of the Tec family tyrosine kinase, interleukin-2-inducible tyrosine kinase (ITK) [1]. ITK is involved in T cell development, differentiation, and effector function, and acts downstream of the T cell receptor (TCR) to mediate phospholipase γ-1 (PLCγ1) phosphorylation and subsequent Ca2+ mobilization.

Given the role of ITK in T cell signalling downstream of the T cell receptor (TCR) and the relevance of this target for inflammatory disorders such as asthma, much medicinal chemistry effort has been applied to the discovery of selective, drug-like ITK inhibitors like GNE-4997. ITK exerts its effects more effectively on T helper 2 (Th2) cells, than on other T cell subtypes, and is crucial for regulating Th2 responses and the development of allergic asthma. No clinical progress has been reported for this compound. |
