Small molecule agents that activate TLR7 and TLR8 and thereby promote pro-inflammatory cytokine release are already in clinical use- for example the TLR7 agonist imiquimod and the mixed TRL7/8 agonist resiquimod. Given that TLR7 and TLR8 are functionally different, selective TLR8 agonists are being sought. |
