GSK-3 negatively regulates several aspects of insulin signalling, and upregulated GSK-3 activity may be involved in the development of insulin resistance in type 2 diabetes [3,7] and in neurodegenerative disease [1,5,9]. GSK-3 inhibitors have therefore become the focus of intense drug discovery efforts. The Wnt signalling pathway is involved in neural stem cell differentiation, mediating the processes of migration [2], synaptogenesis [4], axon guidance [6] and neural induction [10]. GSK-3β inhibition induces decreased activity of the β-catenin degradation complex, thus stabilising β-catenin and mediates increased Wnt signalling. This ultimately leads to increased neurogenesis. |
