Comment: DZX19 was reported as a pan-tropomyosin receptor kinase (TRK) inhibitor [1]. The TRK isozymes A, B and C (encoded by the neurotrophic receptor tyrosine kinase/NTRK genes NTRK1, NTRK2 and NTRK3 respectively) are subject to oncogenic fusions, and are targets for clinically approved anti-tumour drugs such as entrectinib, repotrectinib, taletrectinib and zurletrectinib. DZX19 retains nanomolar inhibitor potency vs. entrectinib-resistant TRKA mutants.
