Compound class:
Synthetic organic
Comment: TYD-68 is a cereblon-recruiting PROTAC that promotes selective degradation of tyrosine kinase 2 (TYK2) [1]. It is a heterobifunctional molecule that binds to the allosteric pseudokinase domain (Janus homology 2: JH2) of TYK2 (using a BMS-986202 derivative) and to cereblon.

Immunopharmacology Comments |
TYK2 is critical for type I interferon, IL-12 and IL-23 cytokine signalling, and is an attractive target for the development of drugs to treat autoimmune diseases. TYD-68 reduces TYK2 protein and downstreat signalling. This action underlies therapeutic efficacy in the imiquimod-induced psoriasis model (mice). |
