Comment: BRP is a bioactive 12 amino acid peptide that is predicted to be cleaved from the BMP/retinoic acid-inducible neural-specific protein 2 (BRINP2) precursor by prohormone convertase 1 (PCSK1) activity [1]. C-terminal amidation (NH₂) and the Leu8 residue are essential for bioactivity. The peptide is proposed to engage a Gα_s-coupled GPCR (in neuronal cells) to mediate its effects on the CREB-FOS signalling pathway (as CREB phosphorylation was sensitive to melittin but insensitive to pertussis toxin and YM254890). The GPCR has not been identified (March 2025).
