Comment: BRP is a bioactive 12 amino acid peptide that is predicted to be cleaved from the BMP/retinoic acid-inducible neural-specific protein 2 (BRINP2) precursor by prohormone convertase 1 (PCSK1) activity [1]. C-terminal amidation (NH2) and the Leu8 residue are essential for bioactivity. The peptide is proposed to engage a Gαs-coupled GPCR (in neuronal cells) to mediate its effects on the CREB-FOS signalling pathway (as CREB phosphorylation was sensitive to melittin but insensitive to pertussis toxin and YM254890). The GPCR has not been identified (March 2025).

Bioactivity Comments
BRP was detected in mouse brain and in human cerebrospinal fluid. In vitro BRP increases Fos expression more potently than glucagon-like peptide 1-(7-37) (GLP-17-37), and activates the cAMP-PKA-CREB-FOS pathway. Administration of the peptide in animal models is anorexigenic and combats obesity, via a mechanism that is independent of leptin, and the GLP-1 and melanocortin 4 receptors [1].
