Comment: Aldumastat (GLPG1972, S201086) is a clinical lead disease-modifying osteoarthritis agent [1]. It inhibits ADAMTS-5-mediated degradation of the cartilage component aggrecan (AGC). This mechanism is predicted to reduce cartilage degradation in inflammed osteoarthritic joints.
