Comment: LEO 134310 is a non-steroidal glucocorticoid receptor (GR) agonist [1]. It was optimised as a topical agent for potential to treat inflammatory skin diseases such as psoriasis. LEO 134310 is rapidly deactivated by enzymatic hydrolysis, thus limiting systemic exposure and improving its therapeutic index. It is hypothesised that LEO 134310 will be more suitable for long-term use than currently used steroidal GR agonist class anti-inflammatory drugs.

In HeLa reporter cell lines the agonist potency (EC50) of LEO 134310 is 1.2 nM for transrepression and 410 nM for transactivation; i.e. a more than 300-fold higher concentration is required to induce transactivation than is needed for transrepression. |
