Comment: DS96432529 was designed as a small molecule, orally active bone anabolic agent, to augment the bone mineralisation effect of osteoporosis drugs such as alendronate, or parathyroid hormone (PTH) [1]. It was selected for its ability to enhance alkaline phosphatase activity, which is an indicator of osteoblast differentiation. Further investigations revealed that DS96432529's pharmacodynamic effects are likely to be mediated via direct inhibition of CDK8 (kinase).
