Comment: PO-322 is an immunosuppressive compound. Its effects are mediated via inhibition of serum/glucocorticoid regulated kinase 1 (SGK1) [1]. SGK1 is a downstream target of mTOR (a significant mediator involved the cell signalling that leads to T cell activation).

Anti‐CD3 or anti‐CD28 treatment stimulates proliferation of human T cells in vitro. PO‐322 inhibits this proliferation (IC50 0.7 μM) [1]. Cytotoxic effects were not evident in vitro. PO‐322 inhibits pro‐inflammatory cytokine production by T cells in vitro, but has no effect on anti‐inflammatory cytokines. It is active in in vivo models, for example reducing imiquimod‐induced dermatitis in mice. |
