Comment: Compound 26 is a macrocyclic inhibitor of apoptosis signal-regulating kinase 1 (ASK1, MAP3K5) [1]. Amongst the analogues designed and tested it is noted for its high level of CNS penetration, making it an ideal lead for examining the effect of ASK1 inhibition on inflammatory demyelination and neurodegeneration. Two previously reported ASK1 inhibitors have shown efficacy in the transgenic mouse SOD1 motor neurone disease model [2].
