Comment SMYD3 is a protein lysine methyltransferase. It was originally described as an H3K4/H4K5 methyltransferase. Subsequent studies revealed that it can methylate nonhistone proteins such as mitogen-activated protein kinase kinase kinase 2 (MAP3K2) [24], vascular endothelial growth factor receptor 1 (VEGFR1) [48], human epidermal growth factor receptor 2 (HER2) [48], and AKT serine/threonine kinase 1 (AKT1) [47], all of which are cancer-related kinases. SMYD3 overexpression may also be oncogenic, and is associated with poor cancer prognosis. Selective SMYD3 inhibition is therefore proposed for anti-tumour potential.
