Immunopharmacology Comments
MDA5 is an intracellualar RNA sensor. It recognizes longer double-stranded RNA sequences than RIG-1. In autoimmunity, MDA5 has been specifically linked with type I diabetes and anti-MDA5 autoantibodies can be detected in patients with certain connective tissue autoimmune conditions [1-3].

Conventional dendritic cells (cDCs), macrophages and fibroblasts isolated from MDA5 null mice have impaired IFN induction after RNA virus infection. Plasmacytoid DCs are unaffected by MDA5 knockout, suggesting nucleic acid sensing TLRs are more relevant in these cells.
