Comment: In >10% of solid tumours aberrant Wnt/β-catenin signalling is a recognised driver of resistance to immunotherapy and is associated with poor prognosis. Zolucatetide (FOG-001; Parabilis Medicines) is a first-in-class peptide that competitively inhibits formation of the β-catenin:T-cell factor (TCF) transcription complex [1]. This action disrupts Wnt signaling, and is proposed for anti-tumour potential. The peptide was created using Parabilis' proprietary Helicon™ technology. Helicons are alpha-helical peptides that are highly modified to optimise target engagement and pharmacokinetics (stability, delivery across the cell membrane). Antibody-Helicon conjugate (AHC) development is the focus of a collaboration between Parabilis and Regeneron that was announced in May 2026.
