Comment: Arctigenin is a lignan class compound that has been isolated from several plant species (incl. Centaurea benedicta/St. Benedict's thistle, Saussurea, Daphne and Trachelospermum species). Several of these plants have been used as traditional herbal remedies for their immunoregulatory, anti-diabetic, anti-tumour, and neuroprotective activities.
Arctigenin has been demonstrated to bind directly to the catalytic site of PDE4D (by X-ray crystallography), and inhibit PDE4D-mediated cAMP hydrolysis at low micromolar concentration (in a scintillation proximity assay) [1].
