RIG-I is an intracellular sensor that responds to viral nucleic acids and activates downstream signaling, resulting in the induction of the type I interferon response [2]. RIG-I recognizes RNAs from more that 25 viruses and can detect dsRNAs, ssRNAs harboring a 5′-triphosphate and blunt-ended 5′-triphosphate-containing dsRNAs. Dysregulated RIG-I signaling has been associated with autoimmune disorders [2].

Conventional dendritic cells (cDCs), macrophages and fibroblasts isolated from RIG-1 null mice have impaired IFN induction after RNA virus infection. Plasmacytoid DCs are unaffected by RIG-1 knockout, suggesting nucleic acid sensing TLRs are more relevant in these cells.
