Poly(I:C) activation of TLR3 is assessed by its ability to induce cytokine production or activation of NF-κB and mitogen-activated protein (MAP) kinases [1].
Immunopharmacology Comments
TLR3 is an endosomal anti-viral receptor [2], with potent pro-inflammatory actions (e.g. induction of Type I IFN and pro-inflammatory cytokines, including IL-6, TNF, IL-17, IL-21, IP-10 and RANTES). Innate Pharma are developing IPH33 as an anti-TLR3 anti-inflammatory biologic, which acts upstream relative to current therapies such as anti-TNF and anti-IL-6 mAbs and small molecule JAK inhibitors.
