High affinity binding of almotriptan to 5-HT1B and 5-HT1D receptors causes constriction of the cranial blood vessels in which these receptors are expressed, which has the effect of inhibiting the release of migraine-causing pro-inflammatory neuropeptides such as calcitonin gene-related peptide (CGRP) and substance P.
