Used in the diagnosis of pheochromocytoma, and in the treatment of hypertensive crisis. Ryzumvi® (formerly Nyxol) is an opthalmic solution containing phentolamine that is indicated for the reversal of pharmacologically-induced mydriasis. When applied topically to the eye phentolamine blocks α1 adrenoceptors within the iris dilator muscle but has no effect on the ciliary muscle.

Antagonism of alpha-adrenoceptors by phentolamine leads to muscle relaxation and widening of blood vessels resulting in a decrease in blood pressure.
